Description: Nervous system and the musculature of the turbellarian Microdalyellia picta (Rhabditophora, Platyhelminthes). The nervous system is stained with antibodies to alpha-tublin. The anterior end of the body showing the brain with the longitudinal nerve cords and the pharyngeal nervous system. The preparation also shows the cilia of the protonephridial terminal cells and the musculature of the body wall and pharynx.

Method: alpha-tubulin, F-actin in muscle fibers (Phalloidin)

Method Description: The worms were fixed for 8 hours with 4% formaldehyde in 0.01M PBS and were kept at 6°С in 0.1% sodium azide in 0.01М PBS. All subsequent procedures were performed at room temperature. Before staining, the turbellarians were processed for 2 hours in 0.5% Triton X-100 in 0.01M PBS and then incubated for 10 hours in antibodies to alpha-tubulin (diluted 1:250, ref. number F2168; Sigma-Aldrich). After incubation, the worms were rinsed for 15 minutes in 0.5% Triton X-100 in 0.01M PBS, stained with phalloidin-TRITC (diluted 1:150, Sigma-Aldrich, ref. number P1951; Sigma-Aldrich) for 2 hours, then rinsed for 15 minutes in 0.01M PBS and mounted in 80% glycerol in 0.01M PBS.

Description: The anterior body end of the turbellarian Microdalyellia picta (Rhabditophora, Platyhelminthes) showing the musculature of the body wall and pharynx and the nervous system stained with antibodies to alpha-tubulin. The series shows the brain, three pairs of longitudinal nerve cords (ventral, lateral and dorsal) that arise from the brain and the pharyngeal nervous system (nerve ring of the juncture line, longitudinal nerves that extend from the ring in the anterior and posterior directions, transverse and circular nerves of the pharynx and neurites running to the receptor cells of the pharyngeal papillae).
